PENETAPAN KADAR TANIN DALAM INFUSA DAUN SALAM (Syzygium polyanthum (Wight.) Walp)) SECARA SPEKTROFOTOMETRI SINAR TAMPAK

ABSTRAK Salah satu tanaman yang digunakan oleh masyarakat sebagai obat tradisional adalah salam (Syzygium polyanthum (Wight.) Walp). Tanin diketahui sebagai salah satu bahan aktif yang terkandung dalam daun salam dan sering digunakan untuk mengatasi masalah diare. Penelitian ini bertujuan untuk mengetahui kadar tanin pada daun salam dengan umur daun yang berbeda. Daun salam muda dan daun salam tua diambil sebagai sampel. Penyarian tanin dari sampel daun dilakukan secara infundasi. Kadar tanin dalam infusa yang diproleh ditetapkan dengan metode spektrofotometri ultraviolet-visibel dengan menggunakan pereaksi Folin Denis dan natrium karbonat jenuh yang akan menghasilkan warna biru. Panjang gelombang maksimum yang digunakan adalah 745,0 nm. Kadar tanin yang diperoleh pada daun salam muda dan daun salam tua berturut-turut sebesar 0,036±1,513% dan 7,07.10-3±0,288%. Dari uji t dengan taraf kepercayaan 95% diperoleh hasil bahwa terdapat perbedaan kadar tanin yang signifikan antara daun salam tua dan muda. Kata Kunci : tanin, daun salam, infusa, spektrofotometri ultra violet-visibel. Abstract People used many traditional medicinal plants. One of them is leaf of salam (Syzygium polyanthum (Wight.) Walp). One of its active compound is tannin that used to treat diarrhoae. The aim of this research was to determine tannin content of salam leaves from different age. Sample that used in this research were young and old salam leaves. Infundation method was used to extract tannin from salam leaves. Tannin content of the infusion was determined by ultraviole-visible spectrophotometry with addition of Folin Denis reagent. Maximum wavelength was 745.0 nm. The result of tannin content in young and old salam leaves were 0.036±1.513% and 7.07x10-3±0.288% respectively. t test showed that there were differences of tannin content in young and old salam leaves.
