PENGARUH PRAPERLAKUAN PEMBERIAN JUS JAMBU BIJI TERHADAP PROFIL FARMAKOKINETIKA TETRASIKLIN PADA TIKUS PUTIH JANTAN

ABSTRAK Penggunaan makanan atau minuman bersamaan dengan obat sering kali dapat menimbulkan interaksi. Tetrasiklin dapat membentuk kompleks tak-larut dengan sediaan besi, alumunium, magnesium dan kalsium, sehingga resorpsinya dari usus gagal. Jambu biji mengandung kalsium 14,00 mg, besi 1,1 mg, karbohidrat 12,20 gram, protein 0,90 gram, dan lemak 0,30 gram, vitamin C lebih dari 490 setiap 100 gram buah jambu biji. Penelitian ini bertujuan untuk membandingkan profil farmakokinetik tetrasiklin dengan praperlakuan 1 jam dan bersamaan serta tanpa perlakuan pemberian jus jambu biji pada tikus putih jantan. Penelitian ini menggunakan rancangan eksperimental sederhana dengan hewan uji tikus jantan galur wistar berbobot 200 g (±10%). Hewan uji dibagi menjadi tiga kelompok, masing-masing kelompok terdiri dari lima ekor tikus. Kelompok 1 diberikan tetrasiklin 63 mg/kg BB. Kelompok 2 diberikan praperlakuan 2 ml jus jambu biji 1 jam sebelum pemberian tetrasiklin 63 mg/kg BB peroral. Kelompok 3 diberikan 2 ml jus jambu biji bersamaan dengan pemberian tetrasiklin 63 mg/kg BB. Setelah hewan uji mendapatkan perlakuan, pada waktu-waktu tertentu diambil cuplikan darah guna penetapan kadar tetrasiklin utuh dengan metode KCKT. Harga-harga parameter farmakokinetika tetrasiklin seperti Ka, Kel, Cpmax, tmax, Cl, t1/2, dan AUC dihitung berdasarkan data kadar tetrasiklin utuh lawan waktu. Parameter farmakokinetika tersebut antar perlakuan dibandingkan secara statistika menggunakan uji ANAVA satu jalan dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan praperlakuan 1 jam sebelum pemberian tetrasiklin dan bersamaan pemberian 2 ml jus jambu biji tidak mempengaruhi parameter farmakokinetika tetrasiklin dosis 63 mg/kg BB yang diberikan peroral pada tikus jantan (P>0,05). Kata kunci: Farmakokinetika,jus jambu biji, tetrasiklin ABSTRACT Drug interaction can happen when consumption food or drink are given together with drug.Tetracyclinecan form insoluble complexes with iron preparations, aluminum, magnesium and calcium, so absorption of intestinal was failure.Every 100 grams of guava fruit contains 14.00 mg of calcium, iron 1.1 mg, 12.20 grams of carbohydrate, protein 0.90 grams and 0.30 grams of fat, vitamin C is more than 490 mg and vitamin A. This research was aimed to comparing the pharmacokinetic profile of tetracycline with pretreatment 1 hour, treated simultaneously and without giving guava juice in male rats.The study was conducted employing a posttest only control group design, using male Wistar rats weight 200 g (±10%). The animals were divided into three groups (5 rats for each group). Group I was given a single oral tetracycline 63 mg/kg BW. Group II was given a single oral of pretreatment 2 ml guava juice 1 hour before treatment with tetracycline 63 mg/kg BW.Group III was given a single oral 2 ml guava juice do simultaneously with tetracycline 63 mg/kg BW. After all rats were pretreated, serial blood samples were withdrawn and were analyzed using HPLC for unchanged tetracycline. Pharmacokinetic parameters of tetracycline i.e. Ka, Kel, Cmax, tmax, Cl, t1/2 and AUC were determined based onconcentration to time data in the blood. The tetracycline pharmacokinetic parameters were analyzed by one-way analysis of varians (ANOVA) using 95% confidence interval.The results showed that the pharmacokinetic parameters of tetracycline in the animal with pretreatment 1 hour and simultaneously of guava juice did notchange significantly (P > 0.05).
