AKTIVITAS FAGOSITOSIS MAKROFAG DARI FRAKSI N-HEKSAN HERBA SAMBILOTO (Andrographis paniculata, (Burm.F) Nees) TERHADAP MENCIT YANG DIINDUKSI VAKSIN HEPATITIS B

ABSTRAK Herba sambiloto (Andrographis paniculata Burm. Nees) mengandung komponen senyawa kimia yang mempunyai aktivitas sebagai imunomodulator. salah satu kandungan zat aktifnya adalah andrographolida yang aktif sebagai hepatoprotektor. Penelitian ini bertujuan untuk mengetahui aktivitas dari A. paniculata sebagai imunomodulator terhadap mencit Balb/c yang telah diinduksi vaksin hepatitis B. Ekstraksi herba sambiloto menggunakan alat Soxhlet dengan pelarut yang berseri. Vaksin hepatitis B digunakan sebagai penginduksi mencit Balb/c. Sebagai tolak ukur imunostimulator adalah peningkatan aktivitas fagositosis makrofag. Hasil penelitian menunjukkan bahwa tiga fraksi aktif dengan peningkatan aktifitas fagositosis makrofag tertinggi sampai terendah yaitu F2, F3, dan F1. Hasil analisis fitokimia menunjukkan kandungan golongan senyawa dari fraksi FI adalah terpenoid, F2 adalah steroid, terpenoid, dan flavonoid; F3 dan F5 adalah terpenoid; serta fraksi F4 mengandung alkaloid. Kata kunci: hepatitis B, herba sambiloto (A. paniculata Burm. Nees), imunomodulator, vaksin. ABSTRACT Andrographis herbs (Andrographis paniculata Burm. Nees) contains many active compounds and showed imunomodulatory activity. One of its component, andrograpolide, has hepatoprotector activity. Therefore, the aim of this study was to find out the immunomodulator compounds from A. paniculata in the BALB/c mice induced by hepatitis B vaccine. Andrographis herbs was extracted using Soxhlet by series of solvents. The isolation process was guided by in vivo imunommodulatory activity in in Balb/c mice induced by hepatitis B vaccine. The imunostimulant activity was assesed through the phagocytic activity of macrophage. The most active extract was fractionated by vaccum column chromatography and further phytochemically identified. The fraction enhanced phagocyte activities of macrophage were F2, F3, and F1. The results of phytochemical analysis showed that the group of compounds from F1 was terpenoid, F2 were steroid, terpenoid, and flavonoid; F3 and F5 were terpenoid; and F4 was alkaloid.
