FORMULASI SEDIAAN TABLET LEPAS LAMBAT ASPIRIN DENGAN ETIL SELULOSA AQUALON T10 SEBAGAI MATRIK

ABSTRAK Aspirin berfungsi sebagai obat analgetik, dapat juga berfungsi sebagai obat antiplatelet. Aspirin sebagai antiplatelet digunakan pada dosis rendah 40 mg/hari. Obat ini memilikiwaktu paro eliminasi yang pendek yaitu sekitar 2-3 jam sehingga harus sering diberikan untuk mempertahankan kadar terapi dalam plasma. Bentuk sediaan lepas lambat dirancang untuk melepaskan suatu dosis tarapetik awal obat yang diikuti oleh suatu pelepasan obat yang lebih lambat dan konstan. Dalam penelitian ini telah dilakukan untuk membuat tablet sediaan lepas lambat aspirin dengan etil selulosa sebagai matrik. Etil selulosa merupakan polimer tidak larut yang bersifat inert yang dapat digunakan sebagai matrik. Penelitian ini bertujuan untuk mengetahui kemampuan etil selulosa dapat digunakan sebagai matrik sediaan lepas lambat aspirin. Sediaan lepas lambat aspirin dibuat dengan metode cetak langsung menggunakan tiga formula konsentrasi etil selulosa yang berbeda yaitu 10%, 15%, dan 20%. Pelepasan aspirin ditentukan melalui uji disolusi dalam medium akuades pada suhu 37° C, dengan kecepatan 30 rpm.Sampel diukur serapannya dengan spektrofotometer UV pada panjang gelombang 265nm. Hasil penelitian menunjukan bahwa semakin besar konsentrasi etil selulosa maka kekerasan tablet semakin meningkat, kerapuhan semakin menurun, dan waktu hancurnya semakin lama. Profil pelepasan aspirin dari matrik etil selulosa mengikuti kinetika orde nol, dengan mekanisme pelepasan difusi. Kata Kunci : aspirin, etil selulosa Aqualon T10, matrik, tablet lepas lambat. Ethyl cellulose is an insoluble polymer that is inert which can be used as the matrix. This study aims to determine the ability of ethyl cellulose can be used as the matrix off slow aspirin dosage. Preparations made with aspirin sustained release off the direct compresion method using three formulas different concentrations of ethyl cellulose is 10%, 15%, and 20%. The release of aspirin was determined through the medium of dissolution test in distilled water at a temperature of 37 ° C, with 30rpm. Sampel velocity measured by UV absorbance at 265nm wavelength. The results showed that the greater the concentration of ethyl cellulose increased the tablet hardness, friability progressively decreasing, and the destruction of the longer time. Release profile of aspirin from ethyl cellulose matrix followed zero order kinetics, with a diffusion release mechanism.
