Aktivitas Sitotoksik dan Ekspresi Protein p53 Bcl-2 Ekstrak dan Fraksi Daun Kersen (Muntingia calabura L.) terhadap Sel Kanker Payudara T47D

Daun kersen (Muntingia calabura L.) merupakan tanaman yang dapat digunakan sebagai alternative pengobatan kanker. Penelitian ini bertujuan mengetahui aktivitas sitotoksik ekstrak etanol daun kersen, fraksi n-heksan, fraksi etil asetat dan fraksi air ekstrak daun kersen terhadap sel T47D dan mengetahui pengaruh ekspresi protein p53 dan Bcl-2 pada pemberian fraksi aktif daun kersen. Daun kersen diekstrak dengan metode maserasi menggunakan pelarut etanol 70%. Ekstrak etanol kemudian difraksinasi dengan partisi cair-cair. Uji sitotoksik dilakukan pada sel kanker payudara T47D dan sel vero dengan metode MTT assay dan dibaca absorbansinya menggunakan ELISA reader. Untuk mengetahui pengaruh ekspresi protein p53 dan Bcl-2 dilakukan uji imunositokimia. Hasil penelitian ini menunjukkan bahwa fraksi n-heksan daun kersen memiliki aktivitas sitotoksik potensial terhadap sel kanker payudara T47D, sedangkan pada ekstrak, fraksi etil asetat, dan fraksi air memiliki aktivitas sitotoksik moderat. Fraksi n-heksan daun kersen mampu meningkatkan ekspresi protein p53 dan Bcl-2 pada konsentrasi 6,79 - 27,16 µg/mL.
